Description

After cultivation, the sample is harvested, and cells are separated from the media using either depth filtration or centrifugation. The next steps depend on whether the recombinant protein is secreted into the media:

Intracellular Proteins/Biologics
If the protein/biologic is not secreted into the media, cell lysis is required to release it. The RPF staff has expertise in various cell lysis techniques, including high pressure, sonication, French press, bead beating, chemical lysis. Lysis is followed by clarification using centrifugation or depth filtration.

Secreted Proteins/Biologics
For secreted proteins or biologics, the clarified supernatant can be concentrated and buffer-exchanged for subsequent purification steps. Our Tangential Flow Filtration (TFF), also called Crossflow filtration system, supports ultrafiltration and microfiltration of samples ranging from 100 mL to over 20 L. These systems include Cytiva, Sartorius, and Merck TFF units with membrane areas of 50, 1,000, and up to 4,500 cm².

    • Two centrifuges 6 x 1L to harvest large volumes of culture

     

    • Lysis by pressure
    • Ideal for larger sample volume

    • Lysis using oscillating ultrasonic waves to lyse cells by cavitation
    • Ideal for smaller sample volume 

    • For hollow fiber filter cartridges and 50 cm2 cassettes
    • Sample volume from 50 to 500 mL

    • For cassettes ranging from  50 cm2 to 0.14 m²
    • Sample volume from 50 to 500 mL

    • For cassettes ranging from 50 cm2 1,000, and up to 4,500 cm².
    • Sample volume from 50 to 20 L
